Efect of hydrogen sulfde donor on oxidative stress in the eye tissues of animals with experimental glaucoma

Studying the features of the glaucoma pathogenesis, considering the high percentage of disability among patients of working age, is an important medical and social problem for society. Adrenaline stress can contribute to the development of regulatory and metabolic disorders, which eventually lead to the glaucomatous process. It is known about the important pathogenetic role of oxidative stress in the development of degenerative diseases of the visual organ, including glaucoma. The probable antioxidant efect of hydrogen sulfde (HS) is being studied. Purpose: To study the efect of eye instillation of 1% sodium hydrosulfde on ophthalmotonus and oxidative stress markers in eye tissues of animals with glaucoma. Material and methods: Glaucoma in rabbits was induced by intravenous injections of adrenaline solution (1:1000) for 3 months. Part of the animals during AIG simulation received daily instillations of 1% sodium HS. Intraocular pressure (IOP) was measured with an applanation tonometer. After the formation of AIG the levels of malondialdehyde (MDA), superoxide radical (SOR) and hydroxide radical (HOR) in the tissues of the drainage zone, retina and optic nerve of the animal eyes was determined. Results: During the formation of AIG in rabbits, a dynamic increase in IOP was observed, which after 3 months of modeling exceeded the norm by 57%. With daily instillations of sodium HS solution, the normalization of the IOP level relative to the initial data by the 60th day was noted. Oxidative stress was observed in the eye tissues of animals with AIG: the content of its markers SOR, HOR and MDA was increased in the tissues of the drainage zone by 92%, 83% and 68%, in the retina by 76%, 68% and 52%, in the optic nerve by 63%, 57% and 38%, respectively, when compared to the control. A lower content of these reactive compounds was observed in the group with instillations of the HS donor. Compared to the data of the AIG group without treatment, this diference was: in the tissues of the drainage zone–53%, 47% and 32%, in the retina–42%, 36% and 24%, in the optic nerve–38%, 34% and 26% respectively. Conclusion: Local administration of HS donor into the eye during glaucoma simulation in rabbits leads to a gradual normalization of increased ophthalmotonus and a decrease in the content of oxidative stress markers in the eye tissues. This may indicate the inclusion of the HS as a gas transmitter in the pathogenetic molecular mechanisms of the glaucoma process.
